Prognostic significance of cyclooxygenase-2 in laryngeal squamous cell carcinoma

Abstract: Epidermal growth factor receptor (EGFR) overexpression is an unfavorable prognostic marker in laryngeal squamous cell carcinoma (SCC). EGFR stimulates cyclooxygenase-2 (COX-2) expression in normal human keratinocytes and squamous carcinoma cells. Based on these observations a prognostic role of COX-2 expression in laryngeal SCC can be hypothesized. Consequently, COX-2 expression was studied in laryngeal SCC (median follow-up = 47 months; range: 2-87 months) by quantitative immunohistochemistry (n = 61) and EGF… Show more

“…These data stand in contrast to a study that demonstrated a correlation between higher levels of COX-2 expression and shorter survival and disease-free survival (14) in a series of laryngeal tumors, indicating that perhaps COX-2 behaves differently in carcinoma of the oropharynx.…”

“…As positive controls for COX-2 antibody, COX-2 positive Hep-2 squamous cancer cells and COX-2 positive squamous cancer tissue specimens (Ranelletti et al, 2001) were always run in the assay. As positive controls for the other antibodies both lymph-nodes and thymus were used.…”

“…The intensity of immunohistochemical staining was evaluated as previously reported (Ranelletti et al, 2001) by the image analysis based on Photoshop (Adobe System, San Jose, CA, USA) together with 'The image processing toolkit' (CRC Press, Boca Raton, FL, USA). The integrated density values (IDV) of the immunostaining was calculated as the product of the mean density value of the immunoreactive regions by the percentage of the immunostained tumour or stroma components.…”

“…Expression of EGFR was markedly increased in the COX-2-transfected cells , and inhibition of COX-2 could suppress the induction of EGFR in these cells [19] . It has also been demonstrated that activation of EGFR can induce COX-2 expression and prostaglandin production [20] . Furthermore, a combination of an EGFR inhibitor with a nonselective COX-1/COX-2 inhibitor was shown to prevent the development of intestinal cancer in nude mice [21] .…”
