Prolonged daily light exposure increases body fat mass through attenuation of brown adipose tissue activity

Abstract: Disruption of circadian rhythmicity is associated with obesity and related disorders, including type 2 diabetes and cardiovascular disease. Specifically, prolonged artificial light exposure associates with obesity in humans, although the underlying mechanism is unclear. Here, we report that increasing the daily hours of light exposure increases body adiposity through attenuation of brown adipose tissue (BAT) activity, a major contributor of energy expenditure. Mice exposed to a prolonged day length of 16-and 2… Show more

“…This allows for metabolic and behavioral processes to be in-tune with 24-hour of day/night cycles (reviewed in Reppert and Weaver, 2002). Disruption of the circadian clock results in arrhythmic physiology and behavior leading to neurodegenerative disorders including Alzheimer’s and Parkinson disease (Videnovic et al, 2014), depression and bipolar disorders (Roybal et al, 2007), learning and memory impairments (Smarr et al, 2014), metabolic syndrome (Kettner et al, 2015), obesity and diabetes (Kooijman et al, 2015), immunodeficiency (Schelermann et al, 2013), accelerated aging (Bunger et al, 2000) and cancer (Altman et al, 2015). About 10% of the genome display circadian expression at the mRNA level in several tissues (Akhtar et al, 2002; Panda et al, 2002; Storch et al, 2002; Ueda et al, 2002; Koike et al, 2012).…”

“…Correspondingly, affecting circadian rhythm by prolonged light exposure in mice reduces sympathetic outflow toward BAT accompanied by decreased BAT activity and increased body fat mass. 28 The importance of the sympathetic nervous system for BAT function is reflected by the number of nerve endings in the tissue. Each brown adipocyte is in close proximity to a nerve ending that releases norepinephrine on sympathetic stimulation.…”

“…Prolonged light exposure reduces the sympathetic stimulation of brown adipose tissue and the ß3-adrenergic intracellular signal. These lower the uptake of fatty acids from triglyceride-rich lipoproteins, and of plasma glucose by brown adipose tissue[43]. …”
