Prolonged Exposure to NMDAR Antagonist Suppresses Inhibitory Synaptic Transmission in Prefrontal Cortex

Abstract: Postmortem studies have shown that schizophrenia produces a reduction in the 67-kilodalton isoform of glutamic acid decarboxylase (GAD67), a key enzyme for gamma-aminobutyric acid (GABA) synthesis. N-methyl-d-aspartate receptor (NMDAR) antagonists have been extensively used to study schizophrenia because they can induce many aspects of the disease, including the decrease in GAD67. It is generally thought that this reduction in GAD implies a reduction in functional inhibition, but direct evidence had been lacki… Show more

“…In vitro studies initially described reduced gamma by ketamine (Cunningham et al, 2006;Zhang et al, 2008), although subsequent studies have refined this description by suggesting a regionally specific ketamine effect such that ketamine increases gamma at the primary auditory cortex but reduces or has no effect in other brain regions (Roopun et al, 2008). Therefore, the observed gamma augmentation at D4 (40-85 Hz) is consistent with prior observations.…”

“…Similar exposures to ketamine in rats lead to an enduring decrease of inhibitory tone in prefrontal cortex (Zhang et al, 2008), supporting the idea that repetitive exposures to NMDA-R antagonists produce enduring effects resembling those observed in schizophrenia.…”

“…The pro-psychotic effects of NMDA-R antagonists appear only in early adulthood and exposure of four week old rats to ketamine on two consecutive days did not reduce GAD67 immunoreactivity (Zhang et al, 2008). However, 24 h continuous exposure of three week old cultured neurons to ketamine produces a substantial reduction in GAD67 immunoreactivity (Kinney et al, 2006).…”

“…Also, these findings exclude a role for disrupted glutamate release in the development of such circuit abnormalities. Indeed, several models suggest disruption of excitatory input into inhibitory neurons as a cause of reduced PV immunoreactivity (60)(61)(62). The shared association between PV cell disruption and the described auditory phenotypes in schizophrenia as well as the Dys1 −/− mice indicates that understanding the mechanisms leading to PV dysregulation may be critical to understanding the development of schizophrenia-associated endophenotypes.…”
