Prolonging photoperiod promotes testosterone synthesis of Leydig cells by directly targeting local melatonin system in rooster testes

Abstract: The study investigated the effects of prolonging photoperiod on the synthesis of testosterone and melatonin in roosters, and the effect of melatonin on testosterone synthesis in rooster Leydig cells as well as its molecular mechanisms. We randomly divided one hundred and twenty 20-week-old roosters into three groups and provided 6, 12.5 and 16 h light, respectively. The results showed that prolonging photoperiod promoted testosterone synthesis, decreased melatonin production, and inhibited the expression of me… Show more

“…Previous studies have shown that melatonin promotes testosterone synthesis in sheep and goat Leydig cells [8,9]. Our previous studies identified the melatonin receptor types of roosters, and our results demonstrated that melatonin could bind to MEL1A and MEL1B to inhibit testosterone synthesis through the cAMP/PKA/CREB signaling pathway in rooster Leydig cells [10]. These results showed that melatonin is species-specific in regulating testosterone synthesis in different species.…”

“…With regard to poultry (such as chicken) ( 40 ), successive digestion of trypsin and collagenase II was carried out in the acquisition of cell suspension. In poultry, collagenase II digestion combined with differential centrifugation was applied in the isolation of rooster LCs, and passage was used in its purification ( 34 ). The contradistinction in digestive enzyme selection is probably owing to the distinct concentration of connective tissues in viviparous and oviparous animals ( 41 ), in which potent trypsin could first digest the connective tissues in chicken testes and then released conglobate seminiferous tubule.…”

“…Currently, the studies related to the isolation and culture of LCs were mainly focused on mice ( 28 ), rats ( 29 ), pigs ( 30 ), sheep ( 31 ), cows ( 32 ), and other mammals ( 33 ). In poultry, collagenase II digestion combined with differential centrifugation was applied in the isolation of rooster LCs, and passage was used in its purification ( 34 ). In contrast, the isolation and identification of highly purified primary LCs in ducks have received very little attention.…”

“…The effects of melatonin on the levels of reproductive hormones depend on physiological conditions and species of animals 35 . Melatonin inhibits StAR, GATA-4/SF-1, or other protein expressions through specific binding sites by blocking StAR protein expression without altering the activity of the P450 enzyme 36 – 38 . Meanwhile, melatonin promotes male reproductive performance and increases testosterone synthesis in mammalian Leydig cells 39 .…”
