Promoter Gene Methylation Regulates Clooxygenase-2 Expression in Androgen-Dependent and Independent Prostate Cancer Cells

Abstract: Background: Clooxygenase-2 (COX-2) expression is overexpressed in human prostate cancer, and aberrant methylation of the COX-2 promoter has also been elucidated. However, how the methylation of CpG islands at COX-2 regulates its expression in prostate cancer is still unclear. We will determine the methylated 5′ CpG island of the COX-2 gene and its role in the expression of COX-2 in prostate androgen-dependent and androgen-independent cancer cells, LNCaP and DU145. Methods:We used western blotting and quantitat… Show more

“…COX-2 is highly expressed in several human cancers and cancer cell lines, including PC tumor cells, and activates the PGE2/EGFR pathway, leading to cell proliferation via extracellular signal-regulated kinase 2 (ERK2) activation. 49 , 50 A study by Zhu et al demonstrated that miRNA-136 suppressed PC cell proliferation and invasion by targeting mitogen-activated protein kinase 4 (MAP2K4a) in vitro 51 . MAP2K4a can increase androgen receptor expression/activation and promote PC tumor progression via noncanonical activation of AKT.…”

“…Peng et al reported the sialidase NEU1 as a biomarker in melanoma [ 17 ]. Liu et al found that promoter gene methylation regulates COX2 expression in prostate cancer [ 18 ], and Kurdi et al found that MRMT-promoter methylation associates with tumor-associated macrophages in advanced astrocytomas [ 19 ].…”
