“…The serum levels of TGF-1 and its mRNA expression level 31 are known to increase in IBD patients. A polymorphism of the TGF-1 gene was found to be associated with stricturing CD 4 and UC 5 . Therefore, this study examined the expression 26 of TGF-1, and demonstrated that poly P treatment downregulated the expression of TGF-1 in both the DSS-or TNBS-induced models of colitis.…”

“…Phosphorylated TGF-1 produced by intestinal epithelial cells and macrophages facilitates the transfer of smad 2/3 and smad 4 from the cytoplasm to the nucleus and promotes the transcription of collagen mRNA in intestinal fibroblasts, thereby inducing the development of intestinal fibrosis [1][2][3] . Recent genome-wide studies have identified a TGF-1 variant to be associated with stricturing CD and UC, suggesting the presence of a strong association 7 between intestinal fibrosis and abnormalities in TGF-1 4,5 . CTGF is associated with wound healing and fibrotic pathology, particularly the induction of sustained fibrosis under cooperation with TGF-1.…”

“…It was postulated that the TT genotype at the -509 position of the TGF-β1 locus was associated with the stricture forming type of the CD (Schulte et al, 2001), and this type of the disease was also associated with a particular genotype at the 915 position (G/C polymorphism) (Hume et al, 2006). Moreover, significant differences in allele frequencies and genotypes at the -800 position (G/A polymorphism) of the TGF-β1 locus were found between patients with UC and control subjects (Tamizifar et al, 2008). However, to our knowledge, all studies on the role of TGF-β1 polymorphisms in IBD reported to date were performed in adult patients.…”

“…Among them, the following four are the most frequent: -800G/A and -509C/T in the promoter region, and 869T/C and 915G/C in exon 1, as shown in Fig 1 (Bayat et al, 2002;Kim et al, 2003;Suzuki et al, 2003;Lee-Chen et al, 2004;Osterreicher et al, 2005). Because of this, and due to the fact that some effects of TGF-β1 polymorphism on IBD was reported to date only for -800G/A, -509C/T and 915G/C (Schulte et al, 2001;Hume et al, 2006;Tamizifar et al, 2008), in our studies we have tested these four polymorphisms.…”

“…A C to G transition at position -509 in the TGFB1 gene promoter has been widely studied as a risk factor for asthma and other inflammatory diseases [13,14]. This polymorphism is located in the silencer region of the TGFB1 gene in a Yin Yang 1 (YY1) consensus binding site, and the presence of the T allele increases basal promoter activity by approximately 30% [15].…”
