The New Jersey strain of EHD virus has been propagated in newborn Swiss mice by the intracerebral route and is regularly lethal beyond the first serial mouse passage. A complement-fixing antigen prepared from the brains of infected mice reacts positively with the sera of deer recovered from infection with either the New Jersey or South Dakota strain of virus, but not with the serum of normal deer. The mouse-passaged virus induced an inapparent infection in an experimental deer. The virus can also be grown serially in HeLa cell culture and induces a characteristic cytopathic effect. It is neutralizable in such cultures to high titer by the sera of deer recovered from EHD (New Jersey strain) and to lower titer by the serum of a deer recovered from EHD (South Dakota strain). Normal deer serum does not neutralize the virus in tissue culture. The HeLa cell-passaged virus induced typical lethal EHD in an experimental deer and virus could be recovered from most of the tissues of this animal in HeLa cell culture. An unexplained prozone of inhibition of cytopathogenicity at low dilutions was observed in cultures of some of the organs. The fact that EHD virus exhibited a limited sensitivity to sodium desoxycholate suggests that it may belong in the arbor virus group.

El virus Junín (JUNV), actualmente clasificado dentro del género Mammarenavirus, es el agente etiológico de la fiebre hemorrágica argentina (FHA). Así como existen algunos mammarenavirus asociados enfermedad humana, también existen cepas de JUNV atenuadas o patogénicas. La cepa de JUNV atenuada mejor caracterizada es la cepa Candid 1 (C#1), de secuencia conocida, y utilizada en la vacuna para prevenir la FHA. En este trabajo se estudió la cepa patogénica P3441 de JUNV, que junto a la cepa C#1 han sido utilizadas en estudios previos donde se ha observado que suscitan distintas respuestas celulares. Con el objetivo de lograr una mayor comprensión de la base molecular de las diferencias observadas entre estas cepas y profundizar nuestro entendimiento de los mecanismos de patogenicidad del JUNV, se obtuvo la secuencia de la cepa P3441 y se realizó un análisis comparativo con otras cepas, incluyendo C#1. Adicionalmente, se clonaron las proteínas virales de la cepa P3441 en un vector bajo control de un promotor procariota para después subclonar cada una de ellas en vectores de expresión bajo un promotor eucariota de forma tal que se expresen unidas a fluoróforos reporteros. Se realizaron estudios de la expresión de las proteínas recombinantes por inmunofluorescencia y microscopía confocal. Posteriormente, con excepción de la proteína L, donde se carece de antisuero, se estudió la reactividad antigénica de las proteínas recombinantes con anticuerpos específicos. Finalmente, a manera de profundizar en la caracterización molecular de la cepa, como JUNV en general, se estudió el sesgo de uso de codones. Los resultados obtenidos y las herramientas generadas sientan la base para profundizar nuestra comprensión de las bases moleculares de la patogenicidad de JUNV.
