Propeptin, a New Inhibitor of Prolyl Endopeptidase Produced by Microbispora. I. Fermentation, Isolation and Biological Properties.

Abstract: Propeptin, an inhibitor of the prolyl endopeptidase isolated from the mycelium of Microbispora sp. SNA-115, is an atypical cyclic peptide antibiotic. It was purified by column chromatographies on silica gel and Sephadex LH-20 and high performance liquid chromatography using an ODS column. Propeptin has the molecular formula of C113H142N26O27 and consists of nineteen amino acids. Propeptin inhibited prolyl endopeptidase of the genus Flavobacterium competitively when Z-Gly-Pro-/?NA was used as a substrate. The i… Show more

“…They form three classes depending on the absence (class II) or presence of one (class III) or two (class I) disulfide bridges that further stabilize the structure [6]. They exhibit a variety of biological activities, involving enzyme inhibition, receptor antagonism, antimicrobial or anti-HIV activities (see Table 1 for class II lasso peptides) [5,[7][8][9][10][11][12]. Their compact structure confers to lasso peptides a great resistance to denaturing conditions and certain proteases.…”

“…nonnitritogenes (IFO14045) and Microbispora indica (IFO14879), but not by Microbispora parva (IFO14043) [8]. Propeptin competitively inhibits the enzyme activity of prolyl endopeptidase (Kiϭ0.7 m M) and also has antimicrobial activity against Mycobacterium phlei IFO3158 (Inhibition zoneϭ12ϳ15 mm at 40 mg/disc) [6,9]. Peptide compounds similar to propeptin have been reported, including anantins (Gly-Asp type) [10,11], RES-701s (Gly-Asp type)…”

“…1(a)) is an atypical cyclic peptide compound that has an amide bond between the a -amino group of the amino-terminal Gly1 and the b -carboxyl group of Asp9, and consists of 16 L configuration amino acids and 3 glycines [6,7]. It is produced by some Microbispora spp., such as Microbispora rosea (IFO14044), Microbispora rosea subsp.…”
