Propylnitrosourea-induced T-lymphomas in LEXF RI strains of rats: genetic analysis

Abstract: Our recent studies have shown that types of lymphomas and leukaemias are determined by host genes in certain experimental models (Yamada et al, 1994a(Yamada et al, , 1994b. Propylnitrosourea (PNU)-induced T-lymphomas are one such example. F344 rats exhibit high susceptibility, whereas many other strains of rats develop predominantly erythro-or myeloid leukaemias (Shisa and Hiai, 1985;Shisa and Suzuki, 1991) We have assumed that a single dominant gene of F344 rats determines the susceptibility to T-lymphomas an… Show more

“…A trove of genetic associations has been established using laboratory rats. For example, studies using RI panels have identified loci that affect cardiovascular disease, 49 hypertension, 50 , 51 diabetes, 52 immunity, 52 tumorigenesis, 53 and tissue-specific gene expression profiles. 54 Using the outbred HS rats, Baud et al.…”

“…A large trove of phenotypic and genomic data have been collected from laboratory rats in the past decades. For example, studies using RI panels have identified loci that control a range of physiological and pathological phenotypes, such as cardiovascular disease, 54 hypertension, 55,56 diabetes, 57 immunity, 57 tumorigenesis, 58 and tissue specific gene expression profiles. 59 Using the outbred HS rats, Baud et al 11 reported 355 QTL in a study that measured 160 phenotypes representing six diseases and risk factors for common diseases.…”

“…DA rats are also susceptible to bladder and tongue carcinomas (Kitano et al 1992), have reduced variation in circadian corticosteroid production (Brodkin et al 1999), and are more easily addicted to morphine (Brodkin et al 1999). F344 rats, on the other hand, are typically resistant to the above conditions, but are susceptible to chemically induced hepatocarcinoma and lymphoma (Lu et al 1999; De Miglio et al 2006) and have decreased bone mineral density (Turner et al 2001). Genetic variation between these strains accounts for most of such strain-specific phenotypes.…”
