Prospective Identification of Enteroviruses Involved in Meningitis in 2006 through Direct Genotyping in Cerebrospinal Fluid

Abstract: Enterovirus infections were investigated with special emphasis on performing rapid molecular identification of enterovirus serotypes responsible for aseptic meningitis directly in cerebrospinal fluid (CSF). Enterovirus genotyping was carried out directly with specimens tested for the diagnostic procedure, using two seminested PCR assays designed to amplify the complete and partial gene sequences encoding the VP1 and VP4/VP2 capsid proteins, respectively. The method was used for identifying the enterovirus sero… Show more

“…EV infections have been known to increase in summer and early autumn in countries in temperate climates ( 1 , 2 , 13 ). As expected in a temperate climate, our surveillance data revealed a seasonal pattern of distribution, with transmission peaking in the summer and decreasing in the period from autumn to spring.…”

“…Laboratory diagnosis of EV infection is based on detection of the virus in clinical specimens such as fecal or rectal swab samples, cerebrospinal fluid (CSF), nasopharyngeal secretions collected by throat swab, and blood ( 11 , 13 ). Detection of EV is usually performed by isolation of the virus in cell culture, reverse transcription PCR (RT-PCR), or real-time RT-PCR ( 11 , 14 – 16 ).…”

“…Viruses were isolated from original clinical specimens by using two cell lines, either MRC5 (human lung embryonic fibroblasts; bioMérieux) or RD (rhabdomyosarcoma; European Collection of Cell Cultures) cell lines. The virus isolates were either typed by seroneutralization test or identified with a genotyping method (Mirand et al, 2008;Nix et al, 2006). A number of virus strains included in the study were subjected to partial sequencing earlier (Diedrich et al, 2009;Huemer et al, 2008).…”

“…A specific protocol was used in one laboratory (Lyon, France) and RNA was purified from 200 ml and eluted in 70 ml. Synthesis of the cDNA was performed using SuperScript III reverse transcriptase (Invitrogen) according to a previously described method (Mirand et al, 2008). …”
