Prospects for cannabinoids as anti‐inflammatory agents

Abstract: The marijuana plant (Cannabis sativa) and preparations derived from it have been used for medicinal purposes for thousands of years. It is likely that the therapeutic benefits of smoked marijuana are due to some combination of its more than 60 cannabinoids and 200-250 non-cannabinoid constituents. Several marijuana constituents, the carboxylic acid metabolites of tetrahydrocannabinol, and synthetic analogs are free of cannabimimetic central nervous system activity, do not produce behavioral changes in humans, … Show more

“…1; ajulemic acid, AJA). The synthesis, chemistry, and biological activity of AJA have been the subjects of several reviews (3,4,24,25). The observation (26) that oral administration of low dose (0.1 mg/kg/day) AJA suppresses joint inflammation and tissue injury in rats with adjuvant arthritis led to further investigation of its mechanisms of action.…”

“…Other reviews cover certain aspects of this subject and the reader is referred to them for a discussion of earlier reports (1)(2)(3)(4). In this review are reports published in the last 5 years on the activities of all classes of cannabinoids, including the endogenous cannabinoids such as anandamide, related compounds such as the elmiric acids (EMAs), and noncannabinoid components of Cannabis that show antiinflammatory action.…”

“…The mechanisms responsible for the anti-inflammatory and joint protective properties of the synthetic cannabinoid AJA are have been studied (Zurier, 2003). Although the elmiric acids are structurally different than AJA, there are certain similarities in their actions so that a comparison of AJA with the elmiric acids may be helpful in understanding both.…”

“…The major active constituent of the plant Cannabis sativa (marijuana), ∆ 9 -tetrahydrocannabinol, and a variety of natural and synthetic cannabinoids have been shown to possess antinociceptive and anti-inflammatory activities (6)(7)(8). For millennia, Cannabis preparations have been used in folk medicine for the treatment of a wide variety of disorders, including those affecting the gastrointestinal tract (9).…”
