Proteasome isoforms exhibit only quantitative differences in cleavage and epitope generation

Abstract: Immunoproteasomes are considered to be optimised to process Ags and to alter the peptide repertoire by generating a qualitatively different set of MHC class I epitopes. Whether the immunoproteasome at the biochemical level, influence the quality rather than the quantity of the immuno-genic peptide pool is still unclear. Here, we quantified the cleavage-site usage by human standard-and immunoproteasomes, and proteasomes from immuno-subunit-deficient mice, as well as the peptides generated from model polypeptide… Show more

“…The T2 cell line is a human T cell leukemia/B cell line hybrid defective in TAP1/ TAP2 (transporter associated with antigen presentation) and ␤1i/␤5i subunits. The T2 cell line contains only standard proteasome (s-proteasomes), whereas the lymphoblastoid cell line possesses proteasomes carrying mainly immuno-subunits, and it is named here as immunoproteasome (i-proteasome) (14,25).…”

“…Two decades of immunoproteasome research solidified the hypothesis of immunoproteasome particles shaping the immunopeptideome presented on MHC class I molecules (Basler et al, 2011;Basler et al, 2006;Basler et al, 2004;Kincaid et al, 2012;Mishto et al, 2014;Morel et al, 2000;Osterloh et al, 2006;Sijts et al, 2000;Toes et al, 2001). Moreover, it became evident that the immunoproteasome has, apart from antigen processing, Fig.…”
