Protection against meningococcal serogroup ACYW disease in complement-deficient individuals vaccinated with the tetravalent meningococcal capsular polysaccharide vaccine

Abstract: SUMMARYIndividuals with properdin, C3 or late complement component deficiency (LCCD) frequently develop meningococcal disease. Vaccination of these persons has been recommended, although reports on efficacy are scarce and not conclusive. We immunized 53 complement-deficient persons, of whom 19 had properdin deficiency, seven a C3 deficiency syndrome and 27 had LCCD with the tetravalent (ACYW) meningococcal capsular polysaccharide vaccine. Serological studies were performed in 43 of them. As controls 25 non-com… Show more

“…In this way, it is quite conceivable that complement deficiency might delay establishment of immunity. By contrast, distinct vaccination responses to polysaccharides have been documented in deficiencies of C3 [13,14] and properdin [14,34], a component of the alternative activation pathway, and might also be expected in classical pathway deficiency states.…”

“…For instance, favorable in vitro responses to tetravalent meningococcal vaccination have been demonstrated in properdin deficiency and in late-complement component deficiencies [10,11,12]. Vaccination of C2D patients has been considered to be advisable despite a lack of supportive data [2,13].…”

“…Complement deficiencies are rare [64], and few studies investigate the impact of these deficiencies on vaccine success. In the largest of these studies, 53 patients with a variety of genetic complement deficiencies (properdin, Factor H, late components) were immunised with the tetravalent meningococcal polysaccharide vaccine ACYW135 [65].…”
