Protective effect of Amphiroa dilatata on ROS induced oxidative damage and MMP expressions in HT1080 cells

Abstract: Antioxidative and matrix metalloproteinase (MMP) inhibitory effects of methanolic extract from ^ãéÜáêç~=Çáä~í~í~ were investigated in HT1080 cells. Treatment with ^K=Çáä~í~í~ extract suppressed the intensity of electron spin resonance signal on the generation of DPPH radicals in a cell-free system and intracellular reactive oxygen species (ROS) formation by 2′,7′-dichlorofluorescin diacetate (DCFH-DA). Radical simulated oxidation of membrane proteins and lipids were also inhibited by treatment with ^K=Çáä~í~í~… Show more

“…UV irradiation is known to provoke oxidative stress through the generation of reactive oxygen species (ROS), which are responsible to interfere with physiological pathways and ultimately activate the overexpression of a number of proteolytic enzymes such as MMPs in skin cells, capable to destroy the dermal connective tissue. In addition, ROS can regulate pro-MMPs activation [11][12][13][14][15] . Thus, the antioxidant activity associated with the inhibition of MMPs could represent a promising strategy to obtain new chemical entities capable to reduce photoaging and prevent wrinkles and damage of the skin, and be effective for other pathological conditions where oxidative stress occurs.…”

“…In accordance with the epidemiological studies, in vivo studies using rodent model demonstrate protective effects of dietary kelp and other red and green algae against mammary carcinogenesis. In East Asian culture, marine algae have always been of particular interest (Khan, Kong, Kim, & Kim, 2010). Edible marine algae accounted for more than 10% of Japanese diet with an average consumption of 1.4 kg per person per year (Burtin, 2003).…”

“…The matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that play important roles in the degradation of extracellular matrix (ECM), and in a variety of biological and pathological processes, such as tumor invasion and metastasis (Sternlicht and Werb, 2001;Nishida et al, 2008;Hwang et al, 2010;Khan et al, 2010). This family of proteinases has includes at least 28 endopeptidases collectively capable of degrading virtually all ECM components.…”
