Tannic acid (TA), a water soluble natural polyphenol with 8 gallic acids groups, is abundantly present in various medicinal plants. Previously TA has been investigated for its antimicrobial and antifungal properties. Being a large polyphenol, TA chelates more than 1 metal. Hence TA has been explored for potent antioxidant activities against reactive oxygen species (ROS), reactive nitrogen species (RNS) and as iron chelator in vitro thereby mitigating iron-overload induced hepatotoxicity in vivo. Iron dextran was injected intraperitoneally in Swiss albino mice to induce iron-overload triggered hepatotoxicity, followed by oral administration of TA for remediation. After treatment, liver, spleen, and blood samples were processed from sacrificed animals. The liver iron, serum ferritin, serum markers, ROS, liver antioxidant status, and liver damage parameters were assessed, followed by histopathology and protein expression studies. Our results show that TA is a prominent ROS and RNS scavenger as well as iron chelator in vitro. It also reversed the ROS levels in vivo and restricted the liver damage parameters as compared to the standard drug, desirox. Moreover, this natural polyphenol exclusively ameliorates the histopathological and fibrotic changes in liver sections reducing the iron-overload, along with chelation of liver iron and normalization of serum ferritin. The protective role of TA against iron-overload induced apoptosis in liver was further supported by changed levels of caspase 3, PARP as well as Bax/BCl-2 ratio. Thus, TA can be envisaged as a better orally administrable iron chelator to reduce iron-overload induced hepatotoxicity through ROS regulation.

Estradiol (E2) is a major determinant of gender-based differences in the development of hepatic fibrosis. MicroRNAs (miRNAs) are endogenous 19-25 nucleotide, noncoding, single-stranded RNAs that regulate gene expression by blocking the translation or decreasing the stability of mRNAs and play an important role in liver fibrosis. The mechanisms underlying the regulation of miRNAs by E2 remain largely unknown. In this study, miR-19b levels were higher and were associated with lower GRB2 mRNA and protein levels in female rats more than in male rats. We also showed that miR-19b levels were down-regulated, were associated with the up-regulation of GRB2 mRNA and protein levels in PS (porcine serum-induced hepatic fibrosis) versus NS (normal control) groups and were up-regulated when associated with the down-regulation of GRB2 mRNA and protein levels in PS + E2 versus PS and in aHSC + E2 (estradiol treated aHSC) versus aHSC groups. MiR-19b expression inhibited cell proliferation in aHSCs, and also down-regulated GRB2 protein expression. The overexpression of miR-19b inhibited cell growth and suppressed COL1A1 protein levels by decreasing the levels of GRB2. However, the forced expression of GRB2 partly rescued the effect of miR-19b in the cells, attenuated cell proliferation, and suppressed the GRB2 protein level by up-regulating the levels of GRB2. Taken together, these findings will shed light on the role of miR-19b in regulating aHSC proliferation via the miR-19b/GRB2 axis. This newly identified miR-19b/GRB2 interaction provided novel insights into the suppressive effect of E2 on HSC proliferation and might facilitate the development of therapies targeting hepatic fibrosis.

This study investigates the seed oils obtained from two different varieties of Opuntia ficus‐indica (Sanguigna and Surfarina) growing in Sicily, Italy. Soxhlet and ultrasound‐assisted extraction procedures are used to obtain the seed oils. The fatty acid profile, γ‐tocopherols, carotenoid content, and the bioactivity in terms of the antioxidant effects (using ABTS, DPPH, FRAP, and β‐carotene bleaching tests), and carbohydrate‐hydrolysing enzyme inhibitory activity (α‐amylase and α‐glucosidase) are determined. The extraction procedure does not significantly affect the fatty acid composition of the oil, but it does affect the tocopherol and carotenoid contents, as well as the antioxidant and inhibitory activities. All the oil samples are rich in polyunsaturated fatty acids, especially linoleic, and oleic acids. Soxhlet extraction proves to be the best extraction procedure to enrich the oils in γ‐tocopherol. Relative antioxidant capacity index (RACI) and global antioxidant score (GAS) analyses are applied to evaluate the antioxidant potential of the Sanguigna variety oil extracted with the Soxhlet apparatus that is shown to be the most active. The same oil also shows the highest α‐amylase inhibitory activity (IC50 value of 40.9 μg mL−1), which is better than that reported for acarbose used as a positive control. Practical Applications: This study is the first to evaluate the effect of oil extraction technologies on the bioactive compound levels and antioxidant and hypoglycaemic activities of cactus pear seed oil, which is a by‐product of fruit processing. The findings will contribute to the valorization of food by‐products. The results provide useful information to select the most valuable technological conditions for oil extraction to preserve the bioactive compounds and the biological activity of the vegetable by‐product extracts, such as seed oil. Cactus pear seed oil obtained by the traditional extraction procedure (Soxhlet) represents a promising source of healthy compounds, particularly of carotenoids and γ‐tocopherol, used not only as antioxidants to preserve lipid components in food preparation but also as functional ingredients due to their hypoglycaemic effect.
