Protective Effects of B2 Preservation Solution in Comparison to a Standard Solution (Histidine-Tryptophan-Ketoglutarate/Bretschneider) in a Model of Isolated Autologous Hemoperfused Porcine Kidney

Abstract: Reperfusion injuries after organ transplantation affect graft function and influence long-term graft survival. As hypothermic storage, which minimizes the extent of unspecific tissue injury after ischemia and reperfusion, is significantly influenced by the composition of preservation solutions, strategies to optimize the different components may lead to longer graft survival. In the present study the effects of the preservation solution B2 on early renal function and histopathological changes were compared to … Show more

“…Then the organs were removed by en bloc technique, arterially cannulated and flushed with preservation solution (4°C) containing 5.000 IE/L heparine (Liquemin N, Roche). 500 ml of preservation solution (see table 2 for B2-solution pursuant to von Baeyer [8]) was then applicated into the artery and the kidneys were transferred under sterile, hypothermic (4°C) conditions from the abattoir to the laboratory.…”

“…Apart from the above mentioned solutions, several other solutions are also utilized for cold storage preservation of kidneys, including phosphate buffered sucrose (PBS) 140 51,52 , HP16 53 , HBS 54 , B2 55 , Lifor 56,57 , Ecosol 58 , Biolasol 59,60 , renal preservation solution 2 (RPS-2) 61 , F-M 62 , AQIXRS-I 63 , WMO-II 64 , CZ-1 65 solutions, etc. Each solution may vary in composition; however, the key strategy with each solution is similar: use colloids, impermeants, electrolytes, antioxidants, and nutrients to minimize ischemic/hypoxic injury and maximize kidney function after reperfusion 66,67 .…”

“…However, as modern ethical standards require reductions of laboratory animal numbers and the use of porcine organs in high numbers is very expensive, an alternative perfusion model was chosen in the present studies that uses organs from slaughterhouse pigs to reduce laboratory animal numbers. This model was developed from existing models of isolated perfused slaughterhouse porcine kidneys (Dittrich et al 2004;Fehrenberg et al 2004;Hochel et al 2003), hearts (Modersohn et al 2001) and skin (GrosseSiestrup et al 2002d), and has been previously established . Also, a valid use for assessing organ toxicity was demonstrated recently (Grosse-Siestrup et al 2002b).…”
