Protective Effects of Clostridium sordellii LT and HT Toxoids against Challenge with Spores in Guinea Pigs.

Abstract: ABSTRACT. The protective effects of Clostridium sordellii lethal toxin (LT) and hemorrhagic toxin (HT) toxoids against challenge with spores in guinea pigs were investigated. Purified LT and partially purified HT were obtained from the culture supernatant of C. sordellii strain 3703, and then were treated with formalin to make toxoids. LT, HT and combined LT and HT (LT / HT) toxoid vaccines were prepared by mixing each toxoid with an aluminum phosphate gel as adjuvant. Guinea pigs immunized twice with the resp… Show more

“…Two C. sordellii virulence factors, lethal toxin (TcsL) and hemorrhagic toxin (TcsH), have also been studied in some detail. Toxoids of TcsL and TcsH are effective vaccines against C. sordellii spore challenge in a guinea pig model (4), indicating that these toxins may play a pivotal role in disease progression.…”

“…A protective antigen, however, does not necessarily equate to an essential virulence factor. Several highly toxigenic TcsL-positive but TcsH-negative isolates have been characterized and shown to be rapidly lethal in an animal model, indicating that TcsH may not be essential for virulence (3,24). As with C. difficile, there are currently no reports documenting the isolation of TcsH-positive, TcsL-negative strains of C. sordellii.…”

“…Gynecological infections in human after childbirth and abortion [147][148][149][150], post-operative infections [4], and necrotizing soft tissue infections associated with heroin use [4,151] can be attributable to infection with C. sordellii. Two toxins are a main responsible virulence factors of C. sordellii: hemorrhagic toxin (TcsH) and lethal toxin (TcsL) [152][153][154][155]. The mortality rate for patients with C. sordellii infection is 40-69%, and most patients die within days or even hours after the initial presentation [4].…”

“…Resembling C. novyi, this infection can be controlled with vaccination [154] in animals, in the production of the toxins at the industrial level, the same problems occur (used later in the formulation of the vaccine). For this reason in our laboratory we had prepared a latex agglutination reagent rapid test for detecting TcsL-toxin of C. sordelli [156].…”
