Protective Effects of Dexmedetomidine and Oxycodone in Patients Undergoing Limb Ischemia-Reperfusion

Abstract: Background: Tourniquet-related complications are a common clinical problem. In the present study, we compared the effects of dexmedetomidine vs. oxycodone in patients undergoing limb ischemia-reperfusion. Material/Methods: Fifty-four patients undergoing unilateral lower-extremity surgery under combined spinal and epidural anesthesia were randomly assigned to a control (ischemia-reperfusion, I/R) group, a dexmedetomidine (Dex) group, and an oxycodone (Oxy) group. Tourniquet-induced hemodynamic parameters change… Show more

“…Both clinically and experimentally, different molecules have been proposed for the prophylactic pharmacological treatment of IRI: (a) antioxidant compounds, such as equivalents of superoxide dismutase (SOD), malondialdehyde (MDA), allopurinol, melatonin, N-acetylcysteine (NAC) [ 12 , 13 , 14 , 15 ]; (b) statins [ 16 , 17 ]; and (c) anesthetics, such as propofol, sevoflurane, dexmedetomidine or oxycodone [ 18 , 19 ]. Another proposed therapeutic approach is the so-called “ischemic conditioning”, first described in myocardium [ 20 ].…”

“…Oxycodone, for instance, has antioxidant and anti-inflammatory properties that protect against surgical stress by inhibiting the production of proinflammatory cytokines and lipid peroxidation. 5 Therefore, it would be of interest to perform a more comprehensive analysis of potential pharmacologic influence on the results.…”
