Protective Effects of Melatonin on Testicular Torsion and Detorsion Damage in Sprague-Dawley Rats

Abstract: SUMMARY:In this study, we evaluated the ultrastructural findings of testis with systemic administration of different doses of melatonin during ischemic period in a rat model of testicular torsion/detorsion (T/D). Testis ischemia-reperfusion (I/R) injury was induced by torsion of the left testis, with a 720 degrees twisting of the spermatic cord so as to produce a total occlusion of testis for 2.5 hours. Subsequently, the same testis was then detorsioned. According to surgical procedure in each group, unilatera… Show more

“…Result of this study show that vitamin C only increase level of testosterone in the damage-induced torsion-detorsion testis. Natural and synthetic antioxidants such as melatonin, hydro alcoholic extract of citrus aurantifolia, coriandrum sativum L seed, quercetin, royal jelly and honey can protect testis from damage-induced by ischemia-reperfusion ( 2 , 39 - 46 ).…”

“…Testicle was fixed in this position for 1 hr by suturing to scrotum ( 14 ). Then, testicle was distorted and placed in the scrotum and gubernaculum was sutured ( 21 ). Rats were then housed individually in cages.…”

“…The Maximal Swimming Time expressed as (min) was measured by forcing the animals to swim against load (5% of body weight) attached about 2 inches from the end of the tail then the animals were held out of the water when become fatigued. For all animals, the body weight, body mass index and abdominal circumference were measured one day before (PTU) and at the end of experiment, blood samples were obtained by decapitation (Aktas et al, 2011) from all rats, the separated sera were stored at -80 o C and used for measurements of the following: Total Tri-iodithyronine (T3), Total Thyroxin (T4) and Thyroid Stimulating Hormone (TSH) by enzyme-linked immunosorbent-assay (ELISA) from Sigma-Aldrich according to the method described by (Das et al, 2008). Triglycerides level was measured by glycerol-3-phosphate oxidase (GPO) enzymatic method obtained from Biodignostic Company (Fossati and Prencipe, 1982).…”
