Protective Effects of Selol Against Sodium Nitroprusside-Induced Cell Death and Oxidative Stress in PC12 Cells

Abstract: Selol is an organic selenitetriglyceride formulation containing selenium at +4 oxidation level that can be effectively incorporated into catalytic sites of of Se-dependent antioxidants. In the present study, the potential antioxidative and cytoprotective effects of Selol against sodium nitroprusside (SNP)-evoked oxidative/nitrosative stress were investigated in PC12 cells and the underlying mechanisms analyzed. Spectrophoto- and spectrofluorimetic methods as well as fluorescence microscopy were used in this st… Show more

“…The cells were disrupted by an ultrasonic processor, and the homogenate was centrifuged at 5000 × g at 4 °C for 15 min to obtain the supernatant for the enzyme activity assays. The enzyme activities of superoxide dismutase (SOD), catalase (CAT), thioredoxin reductase (TrxR), glutathione peroxidase (GPX), glutathione reductase (GR) and glutathione S-transferase (GST) in the Leydig cells were determined according to the methods as previously described 26 , 27 . The enzyme activities of SOD, CAT, GPX and GST were expressed as units/mg protein.…”

“…We did not find pro-inflammatory activity of CeO 2 NPs in HUVE cells. As expected, LPS caused significant activity of both IL-1β and TNF-α when exposed for 24 h. Under normal physiological levels, depending on the cell types and other conditions, NO along with O 2 •− can exert potent anti-inflammatory effects, but NO can also be strongly pro-inflammatory under the condition of its over-production [ 19 ]. NO-donating drugs acting as NOS inhibitors have been reported to cause the release of NO, inhibit IL-1β, and protect against apoptosis in human endothelial and monocytes [ 20 ].…”

“…Rat pheochromocytoma PC12 cells are typically used as models [ 15 ] for studying neuronal degeneration disorders such as Alzheimer’s disease [ 16 ] and Parkinson’s disease. They are also intensively used to investigate the reactive oxygen species (ROS) and NO synthase (NOS) [ 17 ] biochemical pathways involved in cell death and neuroprotection.…”
