Protective effects of thymoquinone and methotrexate on the renal injury in collagen-induced arthritis

Abstract: The goal of this investigation was to study the protective effects of thymoquinone (TQ) and methotrexate (MTX) on collagen-induced arthritis (CIA) in rats. On day 0 under ether anesthesia, the experimental groups were immunized with 0.5 mg native chick collagen II (CII) solubilized in 0.1 M acetic acid and emulsified in Freund's incomplete adjuvant. Control rats were gavaged with vehicle, whereas CII was administered intradermally. In addition, arthritis treated with TQ group received TQ (10 mg kg(-1) bw by ga… Show more

“…This is illustrated by the fact that methotrexate did not produce a significant effect when dosed in a similar therapeutic dosing paradigm even though methotrexate has clear therapeutic benefit in the clinic. It should be noted that methotrexate administered as a prophylactic exhibits significant efficacy in a CIA model (Budancamanak et al, 2006). This profile contrasts with the strong therapeutic effect seen here with dexamethasone, a frank immunosuppressant, and suggests that P2X 7 antagonism may have an effect more similar to methotrexate.…”

“…NC: normal control group; D: disease group; PL: BSO prevention low dose group; PH: BSO prevention high dose group; TL: BSO treatment low dose group; TH: BSO treatment high dose group; MTX: MTX treatment group BSO groups had declining trends of IL-17A, but no effect on IL-4 and IFN-γ expressions. Budancamanak et al (2006) and Tekeoglu et al (2006) reported that TQ had a treatment effect on adjuvant induced or collagen induced arthritis rat models. This observation may be caused by the use of different arthritis models and different medicinal components.…”

“…We suggest that the decrease in GPX activity might be due to partial inhibition of lipid peroxidation and the consequent decrease in the reduction of H 2 O 2 . TQ has been proven to possess a potent antioxidant effect in terms of scavenging properties on free radicals such as superoxide anion [8,9,10,15,38]. A number of studies have demonstrated beneficial effects of various antioxidants and their free radical scavenging property in prostatitis [38,39,40,41].…”

“…The seeds of N. sativa have long been used in the Middle and Far East as a traditional medicine for a wide range of illnesses. TQ is a bioflavonoid with antioxidant, anti-inflammatory, neuroprotective, antiallergenic, antiviral, antidiabetic, and anticarcinogenic properties [8,9,10]. TQ may have a protective effect against inflammation, membrane lipid peroxidation, and infection-induced prostate toxicity.…”
