Protein corona–mediated targeting of nanocarriers to B cells allows redirection of allergic immune responses

Abstract: Opsonization of lectin-coated NPs by activated complement components results in selective B-cell targeting. The intrinsic B-cell targeting property of lectin-coated NPs can be exploited for treatment of allergic immune responses.

“…17) Therefore, the spleen is a good candidate for gene therapy for the treatment of immune-related diseases including allergies, inflammations, infections, and cancer. 20,[92][93][94] The most critical point is to target specific types of spleen cells for achieving a level of gene delivery sufficient for producing a therapeutic effect. This section focuses on in vivo spleen-targeted gene delivery using lipid-based nanocarriers focusing mainly on delivering genes to specific spleen cells.…”

“…117) Colluru et al showed that murine B cells, but not DCs, co-cultured with pDNA efficiently express the antigen and elicit antigen-specific T cell activity in vivo. 118) Shen et al reported that dextran NPs targeted B cells in the spleen through complement activation and produced strong antigen-specific activation. 119) These NPs were used for preventing the induction of anaphylactic shock and allergic asthma.…”

“…The adsorption happens immediately when nanoparticles enter the blood stream and has been termed 'biomolecular or protein corona formation'. [8][9][10] Specific proteins such as immunoglobulins 11 and complement proteins 12 are known to mediate interactions with phagocytic cells and hereby influence the clearance process ('opsonization'). 13,14 There are different reports, which could prove a direct recognition of corona proteins by specific cell-receptors.…”

“…The “protein corona” around NC may affect their organ-specific or cell type-specific trafficking as well as endocytosis and/or functional properties of the NC ( 10 ). Most importantly, the protein corona has been shown to interfere with targeting moieties used to induce receptor-mediated uptake of the NC, both inhibiting ( 11 ) and enhancing ( 12 ) internalization by specific cell types. Moreover, the protein corona is taken up by the target cell, which may alter their function.…”
