Protein corona: Opportunities and challenges

Abstract: In contact with biological fluids diverse type of biomolecules (e.g., proteins) adsorb onto nanoparticles forming protein corona. Surface properties of the coated nanoparticles, in terms of type and amount of associated proteins, dictate their interactions with biological systems and thus biological fate, therapeutic efficiency and toxicity. In this perspective, we will focus on the recent advances and pitfalls in the protein corona field.

“…Protein structural modification and approachability of adsorbed proteins should be carefully considered in the tailoring of preformed protein NPs for effective therapeutic activities mediated through binding with relevant cell receptors. 114 Protein directionality decides the flexibility and availability of optimal binding sites and protein function. Therefore, the directionality of the protein as well as the degree of conjugation/adsorption is essential for NP construction and design in drug delivery.…”

“…The protein corona formation depends on several physicochemical properties of NPs such as size, shape, surface charge, or hydrophobicity and can influence cellular uptake in vitro. [16][17][18][19][20][21][22][23] Therefore, the protein corona can also influence NP functionality and their biological effects. 24,25 For example, transferrin coating was shown to enhance the endocytosis of lipid-coated NPs in A549 cells in vitro and furthermore to have the best inhibitory effect on tumor growth in vivo compared to the other used NPs.…”

“…However, there is no conclusive evidence for the mechanism of SiO 2 NPs inhibition of activated sludge total respiration although indirect evidence from fatty acid methyl ester (FAME) analysis of activated sludge suggests that SiO 2 NPs altered the composition of the microbial membrane lipids [24]. The potential biological effects of SiO 2 NPs on activated sludge warrants further investigations, especially because in nanotoxicity the key role of the interaction of nanoparticles (including SiO 2 NPs) with proteins, which are also a component of activated sludge extracellular polymeric substances (EPS), has been recognized recently [28][29][30].…”
