Protein Disulfide Isomerase-Like Protein 1-1 Controls Endosperm Development through Regulation of the Amount and Composition of Seed Proteins in Rice

Abstract: Protein disulfide isomerase (PDI) is a chaperone protein involved in oxidative protein folding by acting as a catalyst and assisting folding in the endoplasmic reticulum (ER). A genome database search showed that rice contains 19 PDI-like genes. However, their functions are not clearly identified. This paper shows possible functions of rice PDI-like protein 1-1 (PDIL1-1) during seed development. Seeds of the T-DNA insertion PDIL1-1 mutant, PDIL1-1Δ, identified by genomic DNA PCR and western blot analysis, disp… Show more

“…It has been reported that prolyl hydroxylase has chaperone activity and inhibits the aggregation of misfolded proteins that play vital roles in the maturation of secreted plasma membrane and storage proteins (Kim et al, 2012). Prolyl hydroxylase directly provide disulfides to substrate proteins via thiol-disulfide exchange reactions.…”

“…Finally, materials were sectioned into 8 μm thick sections, which were stained with toluidine blue and were observed using a light microscope. Sections were viewed with Olympus BX53 microscope (Olympus, Japan), and were imaged using SPOT FLEX™ camera Whole or sectioned seeds were used for staining with methylene blue [28]. Ten grain seeds were used to measure the aleurone layers thickness.…”

“…Previous studies indicate that PDI mutations can result in irregular starch granule formation and chalky grain phenotypes (206). The rice mutant esp2 lacks protein disulfide isomerase-like (PDIL)1;1, but shows enhanced expression of the thiol disulphide oxidoreductase OsEro1 (207).…”

“…The role of phosphorylation-specific isomerases in protein folding has reported effects on the structure and interactions of seed storage proteins (280,281). Proteins with phosphorylation activity identified with LC-MS/MS, included peptidyl-prolyl cis/trans isomerase (PPIase) and maize protein disulphide isomerase (PDI).…”
