Protein kinase A modulates A‐type potassium currents of larval zebrafish (Danio rerio) white muscle fibres

Abstract: PKA modulates the A-type K(+) current in zebrafish skeletal muscle and affects action potential properties. Our results provide new insights into the role of A-type K(+) channels in muscle physiology.

“…To determine whether PKA is also involved in EGF regulation of activin expression, we tested the effect of a PKA inhibitor (H89) on EGF effects. H89 at 10 lM has been shown to be effective in inhibiting PKA in the zebrafish (Coutts et al, 2009;Garcia-Lopez et al, 2010;Wang and Ge, 2003). Similar to the effects of MAPK3/1, p38 MAPK and PKC pathway inhibitors, the inhibition of PKA activity by H89 also led to a partial reduction of EGF-stimulated expression of inhbaa ($50%).…”

“…In hippocampal pyramidal neurons, activation of PKA by 8-bromo-cAMP significantly decreased I A (36). Similarly, 3-isobutyl-1-methylxanthine stimulation of PKA down-regulated the peak current density of I A (37). In contrast, PKA inducer mimicked the response of serotonin 1D receptor activator and was shown to stimulate I A in TG neurons (38).…”

“…In superficial dorsal horn neurons, stimulation of PKA was found to significantly reduce I A [ 47 ]. Similarly, activation of PKA by forskolin reduced the peak amplitude of I A in rat hippocampal CA1 neurons [ 46 ] as well as in zebrafish white skeletal muscle fibers [ 61 ]. In contrast, a PKA inducer mimicking the effect of neuromedin U receptors, which enhanced I A in dorsal root ganglion neurons, has been reported [ 35 ].…”
