Protein Kinase C-α Activation Induces NF-ĸB-Dependent VCAM-1 Expression in Cultured Human Umbilical Vein Endothelial Cells Treated with Sera from Preeclamptic Patients

Abstract: Aim: To determine if activation of the NF-ĸB-VCAM-1 pathway is mediated by protein kinase C-α (PKC-α). Methods: PKC inhibitor polymyxin B was added to cultured human umbilical vein endothelial cells (HUVECs) from normal pregnancies. Sera from women with uncomplicated pregnancies and with preeclampsia (PE) were added to the control and intervention groups of the HUVECs. Cytoplasmic and membrane PKC, cytoplasmic NF-ĸB inhibitory factor (I-ĸB), and NF-ĸBp65 were measured. Cell viability, cell apoptosis, and VCAM-… Show more

“…PKCb is a predominant conventional isoform of the protein kinase C (PKC) family, and is a serine/threonine protein kinase known to be involved in various cellular signal transductions [13]. Although previous studies linked several members of the PKC family to the pathogenesis of preeclampsia and abnormal placental development [14][15][16][17], very little has been carried out on the specific role of PKCb in preeclampsia. In fact, the regulation of angiogenesis is one of the key roles of these multifunctional isoenzymes in the PKC family, including PKCb [18][19][20].…”

“…Primary HUASMC cultures were established according to the tissue block culture method reported in previous studies. 8 , 11 HUASMC cultures were confirmed by immunofluorescence analysis of the trophoblast marker α-actin, using an anti-α-actin antibody followed by a PE-labeled secondary antibody (both from Santa Cruz, Dallas, TX, USA). Primary HUVEC cultures were established by the trypsin digestion method.…”
