Protein kinase CK2 links extracellular growth factor signaling with the control of p27Kip1 stability in the heart

Abstract: p27(Kip1) (p27) blocks cell proliferation through the inhibition of cyclin-dependent kinase-2 (Cdk2). Despite its robust expression in the heart, little is known about both the function and regulation of p27 in this and other nonproliferative tissues, in which the expression of its main target, cyclin E-Cdk2, is known to be very low. Here we show that angiotensin II, a major cardiac growth factor, induces the proteasomal degradation of p27 through protein kinase CK2-alpha'-dependent phosphorylation. Conversely… Show more

“…Gsk3b is a negative regulator of heart hypertrophy. 33 Interestingly, Hauck 34 recently described that silencing p27 induced cardiomyocyte hypertrophyc growth in the absence of growth-factor stimulation. It is interesting to speculate that Gsk3b mediates negative regulation of hypertrophyc growth through its effects on p27 expression levels.…”

“…The current study highlights an unconventional role for caspases in hypertrophy that does not involve apoptosis. By limiting the duration and degree of caspase activation, CK2 appears central to CT1's nuanced prohypertrophic effects [9], which also involve transient induction of two transcription factors long-recognized to mediate hypertrophy, MEF2 and NFκB. CK2 regulates caspase signaling by phosphorylating both caspase-3 and its targets to inhibit proteolytic cleavage (Figure 1), thus limiting complete execution of the caspase cascade.…”

“…CK2 regulates caspase signaling by phosphorylating both caspase-3 and its targets to inhibit proteolytic cleavage (Figure 1), thus limiting complete execution of the caspase cascade. Of note, CK2 has previously been shown to promote cardiac growth by inhibiting p27, a cell cycle inhibitor [9]. p27 is highly expressed in the heart and downregulated in heart failure.…”

“…The MDC provided expertise in basic science, collaborative opportunities, and an infrastructure that existed nowhere else in Germany. As an example, the clinical departments each and independently produced a keynote paper 15 years after their founding [7][8][9][10]. Detlev Ganten's vision of the Robert-Rössle Clinic and the Franz-Volhard Clinic adjacent to the MDC campus could not survive the expediencies of later times.…”
