Protein palmitoylation and cancer

Abstract: Protein S-palmitoylation is a reversible post-translational modification that alters the localization, stability, and function of hundreds of proteins in the cell. S-palmitoylation is essential for the function of both oncogenes (e.g., NRAS and EGFR) and tumor suppressors (e.g., SCRIB, melanocortin 1 receptor). In mammalian cells, the thioesterification of palmitate to internal cysteine residues is catalyzed by 23 Asp-His-His-Cys (DHHC)-family palmitoyl S-acyltransferases while the removal of palmitate is cata… Show more

“…Palmitoyl‐CoA as an immediate metabolite of dietary PA could also directly be involved in the palmitoylation of an array of proteins, and promote these proteins’ mediated tumor progression . For example, dietary PA could elevate levels of palmitoylated proteins, such as Sonic Hedgehog protein (Shh).…”

“…Additionally, palmitoylation tethers proteins into membrane microdomains such as lipid rafts, thus modulating protein activity, stability, and multiprotein complex formation 4 . Palmitoylation is frequently altered in various diseases including cancer 3,5 . However, the biological role of this PTM is complex and multifaceted and has not been fully investigated due to technical challenges associated with the low abundance of palmitoyl-proteins in general, the difficulty of enriching palmitoyl-proteins with high specificity, and the high hydrophobicity of intact palmitoyl-peptides.…”
