Protein tyrosine kinase 7 is essential for tubular morphogenesis of the Wolffian duct

Abstract: The Wolffian duct, the proximal end of the mesonephric duct, undergoes non-branching morphogenesis to achieve an optimal length and size for sperm maturation. It is important to examine the mechanisms by which the developing mouse Wolffian duct elongates and coils for without proper morphogenesis, male infertility will result. Here we show that highly proliferative epithelial cells divide in a random orientation relative to the elongation axis in the developing Wolffian duct. Convergent extension (CE)-like of … Show more

“…As is the case in other systems (Pfister et al, 2016;Simões Sde et al, 2014;Xu et al, 2016), both cellular intercalation at the luminal surface and cellular migration of cell bodies is dependent on MyoII. Previous work had demonstrated inhibition of cochlear cellular intercalation and junctional remodeling in response to either pharmacological or genetic inhibition of MyoII (Yamamoto et al, 2009).…”

“…PTK7 is an acknowledged regulator of vertebrate PCP and, as such, is involved in various morphogenetic processes (Hayes et al, 2013;Podleschny et al, 2015;Shnitsar and Borchers, 2008;Williams et al, 2014;Xu et al, 2016;Yen et al, 2009). Concerning the functional relevance of the canonical Wnt-mediated endocytosis of PTK7, this could be a mechanism by which a Wnt gradient affects PTK7 localization.…”

“…Once the joining/fusion is complete, the renal vesicle then forms the characteristic J‐/S‐shaped tubules. Therefore, one can hypothesize that a signal promotes the migration of the cells from the renal vesicle toward the Wolffian duct, and elongation may occur via cell proliferation and cell intercalation similar to that observed during Wolffian duct elongation (Xu et al ., ). Interestingly, the epithelium of the short distal region of the mesonephric tubule is similar to that seen in the Wolffian duct (see review by Sainio, ).…”
