Proteomic Analysis of the Hippocampus in Mouse Models of Trigeminal Neuralgia and Inescapable Shock-Induced Depression

Abstract: To investigate the behavioral and biomolecular similarity between neuralgia and depression, a trigeminal neuralgia (TN) mouse model was established by constriction of the infraorbital nerve (CION) to mimic clinical trigeminal neuropathic pain. A mouse learned helplessness (LH) model was developed to investigate inescapable foot-shock-induced psychiatric disorders like depression in humans. Mass spectrometry was used to assess changes in the biomolecules and signaling pathways in the hippocampus from TN or LH m… Show more

“…The mouse TN model was produced by CCI-ION via an intraoral approach as described previously [16]. Briefly, the animal was anesthetized with 4% sodium pentobarbital (50 mg/kg) by intraperitoneal injection.…”

“…The anxiodepressive-like behavioral tests were conducted from day 14 after CION, when the anxiodepressive-like behaviors occurred steadily according to our previous study (30). As shown in Figure 1, D-G, in the anxiety-related behavioral tests, the TN mice showed less time (1-way ANOVA, F 2,30 = 5.96, P = 0.007) and lower entries (1-way ANOVA, F 2,30 = 6.24, P = 0.005) in the open arm of the elevated plus maze (EPM), and they traveled less distance in the center arena of the open field (OF, 1-way ANOVA, F 2,30 = 8.07, P = 0.002).…”

“…Depression is closely associated with abnormal BDNF-TrkB signaling [16,17] together with increased levels of circulating glucocorticoids [18,19] that can be reversed by antidepressants. Disrupted BDNF-TrkB signaling in the hippocampus and the prefrontal cortex has been implicated in the pathophysiology of depression [15,[20][21][22]. In addition, glucocorticoids are released upon stress challenge and regulate metabolic, immune, and neuronal responses [19].…”
