Proteomic analysis of the potato tuber life cycle

Abstract: The tuber of potato (Solanum tuberosum) is commonly used as a model for underground storage organs. In this study, changes in the proteome were followed from tuberization, through tuber development and storage into the sprouting phase. Data interrogation using principal component analysis was able to clearly discriminate between the various stages of the tuber life cycle. Moreover, five well-defined protein expression patterns were found by hierarchical clustering. Altogether 150 proteins showing highly signif… Show more

“…In potato, proteome studies have been carried out mainly on tuber tissue (Bauw et al 2006;Delaplace et al 2005;Lehesranta et al 2005Lehesranta et al , 2006 and in connection with investigation on the action of phytopathogens such as Pectobacterium atrosepticum (Coulthurst et al 2006, Mattinen et al 2007) and the aphid Macrosiphum euphorbiae (Nguyen et al 2007). The application of protein expression studies for cryopreservation is a new research area.…”

“…Gel matching in the present study was based on a reference gel image, created by Progenesis Same Spots (Nonlinear Dynamics) during the image analysis procedure. The reference gel image was used to compare the migration of spots with three previously published potato tuber proteome maps that had been generated by the same 2-DE gel methods to separate protein spots [14,31,32]. Of these three studies, one of them used the same potato variety as used in the present study-Santé [14].…”

“…Of the protein spots whose abundance was significantly (p < 0.01) affected by fertilization and / or previous crop management (PCM) treatments, the function of 47 was inferred by gel matching using 3 published potato tuber proteome maps [14,31,32] and the reference proteome map generated in this study ( Figure 5). Proteins fell into six distinct functional groups: ATP synthesis/binding (2 protein spots), disease/defense/stress response (16 protein spots), energy production-glycolysis (11 protein spots), metabolism (3 protein spots), protein transport/destination/storage (11 protein spots), and protein breakdown (proteolysis/catabolism, 2 protein spots) ( Table 4).…”
