Proteomic analysis of the signaling pathway mediated by the heterotrimeric Gα protein Pga1 of Penicillium chrysogenum

Abstract: BackgroundThe heterotrimeric Gα protein Pga1-mediated signaling pathway regulates the entire developmental program in Penicillium chrysogenum, from spore germination to the formation of conidia. In addition it participates in the regulation of penicillin biosynthesis. We aimed to advance the understanding of this key signaling pathway using a proteomics approach, a powerful tool to identify effectors participating in signal transduction pathways. Results Penicillium chrysogenum mutants with different levels of … Show more

“…The abundance of 628 proteins (26% of the detected proteins) differed significantly in either or both mutants (>400 proteins per genotype). These results highlight the strong impact of protein kinase inactivation on protein production and turnover even in the absence of external stress and is in agreement with other proteomic studies of fungal ST mutants [63][64][65][66][67][68][69] and, in the case of ∆sak1, with the transcriptional results obtained by Heller and colleagues [25].…”

“…Although the key performance indicators Titer, Rate, and Yield (T.R.Y.) have been significantly enhanced in the past 80 years, further improvement is still achievable, for example, using novel tools such as directed evolution, genome sequencing, and “omics” based diagnostics (Carrasco‐Navarro et al, ; Jami, Barreiro, Garcia‐Estrada, & Martin, ; Rokem, Lantz, & Nielsen, ; van den Berg, ; van den Berg et al, ). On an industrial‐scale the fed‐batch strategy is the preferred mode of operation in the production of β‐lactam antibiotics to minimize carbon catabolite repression (Rokem et al, ).…”

“…A traditional fungal global regulator is the heterotrimeric Gα protein Pga1, which is involved in conidiation, sporulation and penicillin biosynthesis [ 107 , 108 , 109 , 110 ]. Carrasco-Navarro and co-workers in 2016 [ 111 ] analyzed several P. chrysogenum Wisconsin 54-1255 mutants with diverse levels of activity of the Pga1-mediated signaling pathway by 2D-DIGE [ 112 , 113 ] (pH 3–10 range and 12% acrylamide SDS-PAGE). The analysis showed how the protein Pga1 is involved in ATP and NADPH control and cysteine biosynthesis, which are required at high levels for penicillin production.…”

“…The analysis showed how the protein Pga1 is involved in ATP and NADPH control and cysteine biosynthesis, which are required at high levels for penicillin production. Thus, a subsequent in silico interactions analysis showed that the protein Pga1 is also involved in morphogenesis, vegetative growth, stress response and penicillin biosynthesis [ 111 ].…”
