Proteomic and Metabolomic Analysis of a Drought Tolerant Soybean Cultivar from Brazilian Savanna

Abstract: Drought is the major abiotic stress which contributes to the reduction in the productivity of soybeans in Brazil, especially in the savanna regions. Two parental genotypes contrasting in drought tolerance (Embrapa 48tolerant and BR 16-sensitive) were used to study the molecular mechanism underlying this process in soybeans. The hydric potential of the BR 16 reached values of −1.0 and −1.5 MPa, three days before the Embrapa 48, confirming the contrasting drought tolerance. The proteomic, phosphoproteomic and me… Show more

“…Soybean seeds from conventional cultivar BR16, considered drought-sensitive (Oya et al 2004;Lima et al 2019) were treated with fungicide Vitavax® Thiram 200 SC (200 g.L −1 ) (ADAPAR) for health quality purposes and then allowed to germinate on Germitest® paper in the dark in germinating chamber, for 4 days, at 28 ± 1°C and 100% relative humidity (RH). Seedlings were transferred to 5 kg pots filled with substrate mixture 1:1 (fertilized soil and washed sand), one plant per pot.…”

“…Aliquots of 50 mg of lyophilized extracts were solubilized in 1.5 ml of cold extraction solution (1 : 2.5 : 1 water, methanol, chloroform) containing 60 μl of ribitol (0.2 mg • ml -1 stock in water) as an internal standard. Metabolic extraction and derivatization were performed following Lima et al (2019).…”

“…In current studies, transcriptomic, proteomic, and metabolomic approaches are mostly used to identify the response of model plants to drought stress ( Chmielewska et al., 2016 ; Michaletti et al., 2018 ; Lima et al., 2019 ; Du et al., 2020 ; Guo et al., 2020 ; Zhou et al., 2022 ), but there is a lack of integrated research on metabolites involved in drought tolerance of forest trees. Therefore, the aim of this study was to measure changes in metabolite levels in the roots and needles of loblolly pine seedlings under long-term drought stress, and to identify major metabolic pathways in loblolly pine seedlings related to drought stress.…”
