Proteomic Characterization of Colorectal Cancer Tissue from Patients Identifies Novel Putative Protein Biomarkers

Abstract: Colorectal cancer (CRC) is one of the leading causes of cancer-related death over the world. There is a great need for biomarkers capable of early detection and as targets for treatment. Differential protein expression was investigated with two-dimensional gel electrophoresis (2D-PAGE) followed by identification with liquid chromatography–tandem mass spectrometry (LC-MS/MS) in CRC patient tissue from (i) the peripheral part of the tumor, (ii) the central part of the tumor as well as from (iii) a non-involved p… Show more

“…GSTπ1 plays a special role in the conjugation of reactive cyclophosphamide metabolites with glutathione [ 63 ]. In the present study, a minor increase (non-significant) in GSTπ1 expression in the descending colon was noted only on D2 in groups ZEN10 and ZEN15 ( Figure 3 ), which suggests that proliferation processes are somewhat dominant in enterocytes [ 64 , 65 ]. These results are difficult to interpret due to the scarcity of published data for comparisons.…”

“…NR1I2 encodes the nuclear receptor Pregnane X, which promotes drug excretion and clearance from the body and mediates the drug-induced upregulation of CYP3A5, and its high expression contributes to the acquisition of resistance in gastrointestinal tumor cells, whereas in vitro and in vitro findings suggest that baicalin significantly inhibits CYP3A5 expression (( Barraclough et al., 2012 ; Ding et al., 2020 ; Noll et al., 2016 ). FABP1 is one of the potential markers of CRC ( Ludvigsen et al., 2021 ; G. L. Zhang, Pan, Huang and Wang, 2019 ). This shows that GQD is closely associated with anti-CRC chemoresistance.…”

“…The raw data files of the EBC samples were searched against the reviewed UniProt Homo sapiens database downloaded 9 February 2020 using MaxQuant (v 1.6.6.0) [27] and used to conduct a relative LFQ analysis of the peptide and protein concentrations. Settings for the database search were as previously described [26] with the match between runs function activated. The LFQ values of each protein were log 2 transformed and technical replicates were averaged by mean.…”

“…LC-MS-analysis was performed on an Orbitrap Fusion Tribrid mass spectrometer coupled online to a Dionex UltiMateTM 3000 RSLC nano system through an EasySpray™ ion source (Thermo Fisher Scientific Instruments, Waltham, MA, USA). LFQ nLC-MS/MS was performed with the universal method settings essentially as described with few modifications [26]. The peptides were loaded onto a 5 mm C18 trap column (PepMap) and separated over a 50 cm C18 main column.…”
