Proteomics study of human cord blood reticulocyte-derived exosomes

Abstract: Reticulocyte-derived exosomes (Rex), extracellular vesicles of endocytic origin, were initially discovered as a cargo-disposal mechanism of obsolete proteins in the maturation of reticulocytes into erythrocytes. In this work, we present the first mass spectrometry-based proteomics of human Rex (HuRex). HuRex were isolated from cultures of human reticulocyte-enriched cord blood using different culture conditions and exosome isolation methods. The newly described proteome consists of 367 proteins, most of them r… Show more

“…Siglec-1 interacts with extracellular vesicles via recognition of sialylated gangliosides packaged on the vesicle membrane [124], which assemble and bud from cellular membranes as viruses do [125,126]. This result has been confirmed not only in vitro [63,127] with extracellular vesicles derived from cell lines or primary cells but also in murine models where Siglec-1 expressed on lymphoid tissues was required to trap extracellular vesicles in vivo [128].…”

“…High resolution multicolor flow cytometry identified three major subpopulations of EVs derived from human plasma High resolution multicolor flow cytometry is one of the recommended methods to quantify EVs from large to small sizes [10]. It allows simultaneous measurement of size and multiple biomarker expression at a single EV level that is not achievable by any other method such as Nanoparticle Tracking Analysis or Transmission Electron Microscopy [15]. Green fluorescent size reference beads with mean diameters of 100, 500, 800, 1000, 3000 and 6000 nm were used to generate a size reference scale ( Fig.…”

“…Human cord blood samples were obtained from the Barcelona Blood and Tissue Bank (www.bancsang.net/), following approval for the protocol and informed consent by the Clinical Research Ethics Committee of Vall d'Hebron University Hospital [PR(CS)236/2017]. Human reticulocyte preparations were performed as previously described (55), with some modifications. Briefly, samples were centrifuged at 1000g for 15 min (acceleration = 6, deceleration = 2), and plasma was removed.…”
