Pseudolaric acid B induces apoptosis via activation of c-Jun N-terminal kinase and caspase-3 in HeLa cells

Abstract: Pseudolaric acid B was isolated from Pseudolarix kaempferi Gordon (Pinaceae) and was evaluated for the anti-cancer effect in HeLa cells. We ob-served that pseudolaric acid B inhibited cell proliferation and induced apoptosis in a time- and dose-dependent manner. HeLa cells treated with pseudolaric acid B showed typical characteristics of apoptosis including the morphological changes and DNA fragmentation. JNK inhibitor, SP600125,markedly inhibited pseudolaric acid B-induced celldeath. In addition, Bcl-2 expres… Show more

“…Cleavage of caspase-3 is a hallmark in the process of apoptosis. Once activated, caspase-3 leads to the final stage of cellular death by proteolytic cleavage of poly (ADP ribose) polymerase (PARP) and DNA on the one hand and activation of pro -apoptotic factors on the other hand (Gong et al, 2004;Kim et al, 2007;Yang et al, 2009). Therefore, we examined the changes in the expression of caspase-3 in PANC-1 cells after exposure to evodiamine, by Western blot analysis.…”

“…The c-Jun/AP-1 transcriptional factor has been of great interest to researchers because its activity is influenced by oxidative stress and can induce apoptosis (Sawai et al, 1995;Gong et al, 2004;Kim et al, 2004). TAJ, also known as TROY, is a -46 kDa membrane protein possessing characteristic cysteine-rich motifs in the extracellular domain and a TNF receptor-associated factor (TRAF)-2 binding sequence in the cytoplasmic domain (Eby et al, 2000).…”

“…However, apoptosis also is implicated in a wide range of pathological conditions, including immunological diseases, allergy and cancer (Holtzman et al, 2000;Zimmermann et al, 2001;Gong et al, 2004;Du et al, 2006). During apoptosis, cells undergo specific morphological and biochemical changes, including cell shrinkage, chromatin condensation, and internucleosomal cleavage of genomic DNA (Reed, 2001;Kankaanranta et al, 2002).…”
