Pseudomonas kitaguniensis sp. nov., a pathogen causing bacterial rot of Welsh onion in Japan

Abstract: Five Gram-reaction-negative, aerobic, motile with one to three polar flagella, rod-shaped bacterial strains, MAFF 212408T, MAFF 212409, MAFF 212410, MAFF 301498 and MAFF 730085, were isolated from diseased Welsh onion (Allium fistulosum L.) in Japan. Analysis of their 16S rRNA gene sequences showed that they belong to the genus Show more

“…Acid production from sucrose, 2-ketogluconate production and margarine hydrolysis were tested using the methods of Lelliott et al [38]. Biochemical and physiological characteristics were also tested using the API 20NE kit (bioMérieux) as in the previous study [26]: the results of the tests were scored after 4 days of incubation at 27 °C, except that nitrate reduction and indole production were analysed after 48 h. In addition, carbon source assimilation tests were conducted using the GEN III MicroPlate system (Biolog) as described previously [26,39]. In brief, the inoculated GEN III MicroPlates were incubated at 27 °C for 4 days and read with the Biolog Micro-Station semi-automated reader; the results were interpreted using the MicroLog 3 software (Biolog).…”

“…Phylogenetic analysis based on the 16S rRNA gene sequences was conducted in the same manner as in our previous study [26], targeting the broccoli isolates and the type strains of the top 31 Pseudomonas species found to be close to the isolates based on the 16S rRNA gene sequence similarities. Briefly, the sequences of the broccoli isolates, the top 31 species and Pseudomonas aeruginosa JCM 5962 T selected as an outgroup were subjected to phylogenetic analysis using mega X [27].…”

“…Phylogenetic analysis based on the 16S rRNA gene sequences was conducted in the same manner as described in our previous studies [7, 8], by targeting the five cyclamen isolates and the type strains of the top 30 Pseudomonas species, which were found to be close to the isolates based on the 16S rRNA gene sequence similarities (). Briefly, the sequences of the cyclamen isolates, the top 30 species, and Pseudomonas aeruginosa JCM 5962 T , selected as an outgroup, were subjected to analysis using mega X ver.…”
