Psychopharmacology: From serendipitous discoveries to rationale design, but what next?

Abstract: Psychopharmacology really developed as a discipline from the mid-20th century with the discovery of a number of new classes of psychoactive drugs which could modify behaviour. These drugs were discovered as a consequence of clinical observations of patients, often being treated for other conditions. These serendipitous discoveries were the start of an era of drug development which has led to the antidepressants, antipsychotics, anxiolytics and mood stabilisers used today. Subsequent research focused on underst… Show more

“…This is due to both the high prevalence of MDD and its frequent comorbidity with other conditions (2)(3)(4), particularly cardiovascular disease (5). Most antidepressants target monoamine-related pathways (6) but are ineffective in 40% of cases (7). Better understanding of the molecular mechanisms of MDD may aid in the development of more effective treatments.…”

“…After the discovery of the antidepressants, antipsychotics, anxiolytics and mood stabilizers currently in use today, research slowed down and most major pharmaceutical companies decreased their investment in psychopharmacology. However, new avenues are now being explored [ 107 , 108 ]. Renewed interest in this area and research on the NPY system may eventually bring the expected benefits for human health by drugs interfering with this system.…”

“…Most studies have addressed their pharmacological profiles only after acute treatment in males, whereas the efficacy and side-effect profile of these compounds after chronic treatments remain uncertain. Various drugs used in the treatment of psychiatric disorders cause side effects, tolerance and withdrawal symptoms after long-term use (Guina and Merrill, 2018;Robinson, 2018;Krause et al, 2019). Thus, studies addressing the effects of chronic administration of TRPV1 blockers are warranted.…”
