Punicalagin Pretreatment Attenuates Myocardial Ischemia-Reperfusion Injury via Activation of AMPK

Abstract: Punicalagin (PUN), a major bioactive component in pomegranate juice, has been proven to exert neuroprotective effects against cerebral ischemia/reperfusion (I/R) insult via anti-oxidant properties. This study aims to investigate whether PUN provides cardioprotection against myocardial I/R (MI/R) injury and the underlying mechanisms. PUN (30 mg/ kg/d) or vehicle was intragastrically administered to Sprague-Dawley rats for one week before the operation. MI/R was induced by ligating the left anterior descending c… Show more

“…Rat hearts and cardiomyocytes were lysed with RIPA buffer (P0013; Beyotime Biotechnology) containing protease inhibitor cocktail. Western blotting was carried out utilizing the standard method as described previously . The primary antibodies used were as follows: Drp1 (#14647; Cell Signaling), Fis1 (#GTX111010; Genetex), Opa1 (#612606; BD biosciences), Mfn2 (#9482; Cell Signaling), Mfn1 (#GTX133351; Genetex) and β‐tubulin (#66240‐1‐Ig; Proteintech).…”

“…Melatonin, a compound isolated from Lycium barbarum L. (Gouqizi), can exert a cardioprotective effect via the AMPK-PGC-1a-SIRT3 signaling pathway (Yu et al, 2017). Punicalagin, a bioactive component isolated from Punica granatum L. (Shiliupi), was reported to exert cardioprotective effects by restricting oxidative stress and myocardial injury in a rat model of MI/R via activation of the AMPK signaling pathway (Ding et al, 2017). Berberine is a component extracted from Phellodendron chinense C.K.Schneid.…”
