Purification and characterization of a neurotoxic phospholipase A2 from Indian cobra (Naja naja naja) venom

Abstract: Snake venoms contain multimolecular forms of phospholipase A2 which are diverse with respect to their pharmacological properties. A neurotoxic PLA2 from Naja naja naja venom has been purified in two steps. (1) The whole venom was fractionated on CM-Sephadex C-25 column; 4.6% of the total PLA2 activity recovered was found in the NN-V fraction. (2) The NN-Vb-PLA2 fraction was purified to homogeneity by gel filtration of fraction NN-V on Sephadex G-50. It is a basic protein with a mol. wt between 10,500 and 11,00… Show more

“…whereas there was only one (minor) neutral phospholipase A 2 (peak 3) in the non-spitting N. kaouthia venom. Basic phospholipases A 2 are known to be involved in many pharmacological actions of cobra venoms including myotoxicity, cytotoxicity, cardiotoxicity and edema-inducing activities (23)(24)(25)(26)(27)(28). Also, we note that all three Southeast Asian spitting cobra venoms contained a substantial amount (30-40%) of polypeptide cardiotoxins (Table 3).…”

“…The alkaloid also inhibits the edema inducing activity of TFV PL-X but on the other hand it enhances the edema inducing activity ofTFV PL-la and TFV PL-lb PLA2 enzymes [23]. Further two neurotoxic PLA2s, NNIVb-PLA 2 and NN-Vb-PLA2 devoid of edema inducing activity have been isolated from Naja naja naja venom [24,25]. Chlorpromazine failed to inhibit the edema inducing activity of VRV-PL-VIIIa enzyme indicating that the edema inducing property may not be always related to the PLA2 activity and different mechanisms may be involved in the edema inducing activity of PLA 2 enzymes.…”

“…This experiment was carried out by employing the method described in the previous report Gowda, 1989 andBhat et al, 1991) which involves the measurements of free fatty acids liberated by the enzyme catalyzed hydrolysis of egg-phosphotidyl choline. A set of 7 cleaned and dried vials were taken; to this, 50µl of phosphotidyl choline and 50-200ug of concentrated venom samples were added.…”
