Purification and characterization of an immunosuppressive factor from ovarian cancer ascites fluid

Abstract: A nonspecific immunosuppressive factor present in malignant (ovarian carcinoma) ascites fluid has been purified by acid extraction from a high molecular weight (greater than 20000) complex followed by preparative isoelectric focusing on Bio-lyte media. It is an acidic protein (pI = 3.6) of mol. wt. 50000 to 52000 as estimated by gel filtration and composed of subunits of 25000 to 26000 estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis, under reducing conditions. It inhibits the phytohemagg… Show more

“…A variety of reports have documented the presence of immunosuppressive humoral factors present in the serum or ascitic fluid of cancer patients. A number of these have described an acidic proteineic factor present in the ascites of ovarian carcinoma patients [3,25,34,39]. Characterization of this substance revealed a protein of 50-52 kDa molecular mass composed of subunits of 25-26 kDa with the ability to inhibit spontaneous cytotoxicity ofperipheral blood lymphocytes (PBL) against K562 cells.…”

“…In order to avert this effect, purification of immunoprotective TSTA free of tumor-enhancing substances was achieved by pIEF of 3 M KC1 extracts to separate the active material from acidic fractions which facilitate tumor growth in vivo by activating suppressor cells [10,26]. Similar acidic immunosuppressive factors have been described for human cancers [10,18,33]. In contrast to 3 M KC1 extracts, butanol extracts and electrofocused fractions of butanol extracts do not enhance tumor growth [13].…”

“…To the best of our knowledge, no information is available about the PHA-induced expression of HLA Class II antigens and the functional activity in autologous MLR of T cells isolated from draining lymph nodes (LN-T) from patients with solid tumors. In view of the release of immunoregulatory factors from human tumors [10,14,19,20,26], in this study we have compared the induction of HLA Class II antigens and the functional properties in autologous and allogeneic MLR of T cells isolated from mediastinal lymph nodes and from peripheral blood of patients with lung carcinoma.…”
