Purification and characterization of an extracellular chitinase from antagonistic Streptomyces violaceusniger

Abstract: The actinomycetes Streptomyces violaceusniger showed strong antagonistic activity against various tested wood rotting fungi. An extracellular chitinase, produced by antagonistic S. violaceusniger MTCC 3959, was purified as follows: ammonium sulfate precipitation, chitin affinity and chromatographic separation of Q Sepharose. The molecular mass of the purified chitinase was estimated as 56.5 kDa by SDS-PAGE. Chitinase was optimally active at pH of 5.0 and 50 °C. It retained almost 100% activity at pH 5.0 and al… Show more

“…The major inhibitor of chitinase activity was Hg 2+ since it reacts with -SH groups found in cysteine residues in the protein chain and disrupts the tertiary structure. It strongly inhibits chitinases from different genera including Bacillus (Waghmare and Ghosh, 2010), Massilia timonae (Adrangi et al, 2010), Microbispora (Nawani et al, 2002), Rhodothermus (Hobel et al, 2005), Stenotrophomonas (Jankiewicz et al, 2012) and Streptomyces (Nagpure and Gupta, 2013;Prakash et al, 2013). The other major inhibitory metal ions which show strong inhibitory action on chitinases include Fe 2+ (Pradeep et al, 2014;Wang et al, 2010), Mn 2+ (Pradeep et al, 2014;Wang et al, 2010), Ag 2+ (Nagpure and Gupta, 2013;Liu et al, 2010), Zn 2+ (Jankiewicz et al, 2012;Prakash et al, 2013) and Co 2+ (Jiang et al, 2012;Yong et al, 2005).…”

“…Among them, we included important substrates from plant and fungal origins, such as microcrystalline cellulose (Serva), carboxymethyl cellulose (Sigma-Aldrich), xylan from beechwood (Sigma-Aldrich), starch (Sigma-Aldrich), and colloidal chitin prepared according to Nagpure and Gupta (51). The flasks were incubated at 24°C with shaking at 300 rpm for 7 days.…”

“…A Lineweaver-Burk plot was obtained by plotting 1/v against 1/s. Kinetic parameters (Km and Vmax) were estimated by linear regression from Lineweaver-Burk plot (Nagpure and Gupta, 2013;Rahman et al, 2014).…”
