Purification and Characterization of Levansucrases fromBacillus amyloliquefaciensin Intra- and Extracellular Forms Useful for the Synthesis of Levan and Fructooligosaccharides

Abstract: The intra-and extracellular levansucrase (LS) activities produced by Bacillus amyloliquefaciens were promoted by supplementing the sucrose medium with yeast and peptone as nitrogen sources. These activities were purified by polyethylene glycol (PEG) fractionation for the first time. PEGs of low molecular weight selectively fractionated the intracellular LS activity rather than the extracellular LS activity. Contrary to other LSs, B. amyloliquefaciens LSs exhibited high levan-forming activity over a wide range … Show more

“…Bacillus circulans (Perez Oseguera et al 1996), Bacillus amyloliquefaciens (Tian et al 2011), Bacillus sp. , and Geobacillus stearothermophilus (Inthanavong et al 2013) require sucrose as inducer to improve the levansucrase production and a very low or even no significant levansucrase production is achieved in the absence of sucrose.…”

“…B. amyloliquefaciens (Tian et al 2011), B. subtilis NRC33a (Abdel-Fattah et al 2005, and Bacillus sp. prefer to utilizing yeast extract to produce levansucrase, by contrast, high level of levansucrase from G. stearothermophilus is produced in the medium comprised of peptone or tryptone than yeast extract (Inthanavong et al 2013).…”
