Purification and Characterization of the Human Interleukin-18 Receptor

Abstract: Interleukin (IL)-18 was identified as a molecule that induces IFN-␥ production and enhances NK cell cytotoxicity. In this paper, we report upon the purification and characterization of human IL-18 receptor (hIL-18R). We selected the Hodgkin's disease cell line, L428, as the most strongly hIL-18R-expressing cell line based on the results of binding assays. Murine interleukin-18 (mIL-18) 1 was identified in the livers of mice sequentially injected with heat-killed Propionibacterium acnes and with lipopolysacchar… Show more

“…Initiation of intracellular signalling by secreted IL-18 starts with the formation of a high-affinity receptor complex, comprising out of the IL-18 binding areceptor chain (IL-18Ra) and the b-chain (IL-18Rb), which itself cannot bind IL-18. 10,11 Dimerization of both receptor units upon IL-18 binding results in activation of intracellular signal transduction pathways induced by recruitment of the adapter protein MyD88 towards the receptor complex, followed by binding of interleukin receptor activated kinase (IRAK) and TNF receptor associated factor-6 (TRAF-6). The formation of this activation complex of the IL-18 signal cascade finally leads to downstream NFkB-mediated activation of transcription.…”

“…It also plays a role in the development and activation of Th1 cells but not Th2 cells. 10 Although the receptor of IL-18 has been recently identified, 11 its biological significance is under investigation. In this study, we tested whether the cytokines, which act on Th1 cells, can be used for cancer gene therapy.…”

“…This is reminiscent of IL-1 signaling, which involves both IL-1R type I and IL-1R accessory protein (IL-1RAcP) (8,9). Gene-targeting studies have revealed that IL-18R␣ Ϫ/Ϫ mice are defective in IL-18-mediated biological functions (6,10). However, the physiological functions of AcPL remain to be investigated.…”
