Purification and identification of novel cytotoxic oligopeptides from soft coral Sarcophyton glaucum

Abstract: Globally, peptide-based anticancer therapies have drawn much attention. Marine organisms are a reservoir of anticancer peptides that await discovery. In this study, we aimed to identify cytotoxic oligopeptides from Sarcophyton glaucum. Peptides were purified from among the S. glaucum hydrolysates produced by alcalase, chymotrypsin, papain, and trypsin, guided by a methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay on the human cervical cancer (HeLa) cell line for cytotoxicity evaluation. Purification tech… Show more

“…These compounds manifested low cytotoxicity on non-cancerous Hek293 cells (Supplementary Table 1). Besides that, AERQ, RDTQ and AGAPGG have cytotoxic activity that is 5.8, 5.1, and 3.3 times stronger than that of the anticancer drug 5-fluorouracil, respectively (Quah et al, 2019).…”

“…Hydrostatin-SN1 was proposed to be a promising candidate for the development of treatments for acute lung injury [ 164 ] and inflammatory bowel diseases [ 371 ]. Bioactive peptides were also reported from the giant barrel sponge, Xestospongia testudinaria [ 373 ] and rough leather coral, Sarcophyton glaucum [ 374 ] KENPVLSLVNGMF derived from X. testudinaria was dose-dependently cytotoxic to the human cervical cancer cell line (HeLa), being 3.8-fold stronger than anticancer drug 5-fluorouracil. By contrast, the peptide showed only a marginal 5% cytotoxicity to Hek293, a non-cancerous, human embryonic kidney cell line [ 374 ].…”

“…They displayed relatively high cytotoxicity on HeLa cells (human cervical cancer cells), which was 3.3-, 5.8-, and 5.1-fold stronger than that of the anticancer drug 5-FU, respectively. Additionally, their IC 50 values to inhibit the growth of Hela cells were 8.6, 4.9, and 5.6 mmol/L, respectively [ 41 ].…”
