Purification, properties and primary structure of thioredoxin from Aspergillus nidulans

Abstract: This paper reports the purification and the properties of a thioredoxin from the fungus Aspergillus nidulans. This thioredoxin is an acidic protein which exhibits an unusual fluorescence emission spectrum. characterized by a high contribution of tyrosine residues. Thioredoxin from A . nidulans cannot serve as a substrate for Escherichia coli thioredoxin reductase. Corn NADP-malate dehydrogenase is activated by this thioredoxin in the presence of dithiothreitol, while fructose-I ,6-bisphosphatase is not. The am… Show more

“…S1A). AnTrxA exhibits all the characteristics of thioredoxins and represents the A. nidulans thioredoxin sequence (accession number AAB24444) described earlier (32). Here, by reverse transcription of the gene designated with accession number XM_656093, we identified a shorter cDNA version for the A. nidulans thioredoxin reductase (accession number AM396558).…”

“…Numerous thioredoxin amino acid sequences are found in the literature (e.g. [17]) and their comparison shows that thioredoxin f is not very similar to other thioredoxins (30 ~o similarity), except for the conserved active site sequence. This is shown indirectly by the fact that the thioredoxin f antibodies do not recognise any of the other thioredoxins tested.…”
