Pyomelanin-Producing Brevundimonas vitisensis sp. nov., Isolated From Grape (Vitis vinifera L.)

Abstract: A novel endophytic bacterial strain, designated GR-TSA-9T, was isolated from surface-sterilized grape (Vitis vinifera L.). 16S rRNA gene sequence analyses showed that the isolate was grouped within the genus Brevundimonas, displaying the highest similarity with Brevundimonas lenta DS-18T (97.9%) and Brevundimonas kwangchunensis KSL-102T (97.8%) and less than 97.5% similarity with other members of Brevundimonas. The strain GR-TSA-9T was a gram negative, rod shaped, facultatively anaerobic, catalase and oxidase … Show more

“…Brevundimonas have been utilised in soil bioremediation (Zhang et al 2021 ) and in treating polluted water (Wang et al 2015 ). Recently, Jiang et al ( 2021 ) reported the isolation of a novel pyomelanin-producing B . vitisensis from a grape; however, this is the first study to report the isolation of a novel red pigment from Brevundimonas .…”

“…The growth of strain DM2-R-LB4 T was observed in different media, namely LBA, marine agar 2216 (MA, Difco), malt extract agar (MEA, Difco), potato dextrose agar (PDA, Difco), Reasoner's 2A agar (R2A, MB cell), nutrient agar (NA, Difco), ISP2 (yeast extract 4.0 g l -1 , malt extract 10.0 g l -1 , dextrose 4.0 g l -1 , agar 20.0 g l -1 ) and trypticase soy agar (TSA, Difco). Growth on TSA medium was observed at different temperatures (4,10,15,20,25,30,35,37,40,45, 50 and 55 °C) and different pH (pH 4.0-10.0, at intervals of 0.5 pH unit). Two separate buffers were used for studying growth at different values of pH.…”

“…Polyamines were extracted from the freeze-dried cells and analysed as described by Busse and Auling [34]. The polar lipids were extracted from 100 mg of freeze-dried cells according to the method described by Jiang et al [35]. The polar lipids were separated by two-dimensional thin-layer chromatography on 20×20 cm silica gel plates (Merck) and identified by spraying with phosphomolybdic acid, molybdenum blue, and ninhydrin [36].…”

“…This research highlights the potential use of thermotolerance genes and SM-BCGs to enhance stress tolerance and engineer crops resistant to various challenges. For example, previous studies explored bacterial strains producing pyomelanin, which aids grapes in preventing UV damage (Jiang et al, 2021), and nostoxanthin, known to reduce reactive oxygen species and improve salt stress (Jiang et al, 2023). These findings indicate that the BRMEA1T strain might play a significant role in facilitating thermotolerance adaptation in host plants.…”
