QseC Mediates Osmotic Stress Resistance and Biofilm Formation in Haemophilus parasuis

Abstract: Haemophilus parasuis is known as a commensal organism discovered in the upper respiratory tract of swine where the pathogenic bacteria survive in various adverse environmental stress. QseC, a histidine protein kinase of the two-component regulatory systems CheY/QseC, is involved in the environmental adaptation in bacteria. To investigate the role of QseC in coping with the adverse environment stresses and survive in the host, we constructed a qseC mutant of H. parasuis serovar 13 strain (ΔqseC), MY1902. In thi… Show more

“…Bacteria must source sufficient iron from the host and maintain a dynamic balance for their growth. Iron plays a key role in the growth of H. parasuis, and low availability in a host is a primary pressure for invasion of pathogenic bacteria and is considered a signal that leads to significant changes in cell processes (Deslandes et al, 2007;He et al, 2018). In the present study, 100 µM BIP was added to fresh T/V/S medium to construct an iron-restricted environment to analyze the capability of HPS5 and crp to utilize iron.…”

“…The ability of wild-type and crp mutant strains to utilize iron was measured as previously described (He et al, 2018;Dong et al, 2019) with some modifications. Briefly, the two strains were cultured to log-phase replication, and the value of OD 600 of the cultures was adjusted to 0.4.…”

“…AI-2 has either a positive or negative effect on biofilm formation in different bacteria. 3/ AI-3 mainly in Gram-positive systems typically use secreted oligopeptides [13,14] and two-component systems (TCS), which consist of membrane-bound sensor kinase receptors (QseC) and cytoplasmic transcription factors (QseB) which direct alterations in gene expression [2,15]. The QseC receptor responds to AI-3 but also to AI-2 and, additionally, to epinephrine/ norepinephrine from eukaryotic hosts.…”

“…The biofilm formation test was performed as previously reported with some modifications [29,30]. Briefly, 20 µL of overnight culture of the wild-type strain and its mutants were inoculated into 2 mL of fresh TSB broth containing NAD and inactivated serum with or without glucose in 6-well plates, and then incubated for 24 h or 72 h at 37 °C.…”
