QTc interval prolongation in patients with systemic lupus erythematosus treated with hydroxychloroquine

Abstract: Objectives: The primary objective is to reveal the effect of hydroxychloroquine (HCQ) treatment on corrected QT (QTc) interval in patients with systemic lupus erythematosus (SLE). The secondary objective is to investigate factors that affect QTc prolongation. Methods: SLE patients who had electrocardiograms between 2015 and 2020 were recruited and assigned to two groups based on whether they were treated with HCQ (HCQ group) or not (control group). Change of QTc before and after HCQ administration in the HCQ gr… Show more

“…Despite this finding, case reports for TdP and sudden death have been described for hydroxychloroquine when used for non-malaria indications [ 35 ]. In a Japanese study on systemic lupus erythematosus patients treated with hydroxychloroquine, a significant increase on the QTc was found when compared with a control group with no hydroxychloroquine exposure [ 36 ].…”

“…In a separate cohort study conducted by Nishiyama et al, the primary objective was to evaluate the impact of HCQ treatment on QTc interval among patients diagnosed with SLE during the period spanning from 2015 to 2020 23 . In addition to this primary goal, the study also aimed to identify various factors that could potentially influence QTc prolongation.…”

“…Furthermore, the impact of HCQ treatment on QTc interval in patients with SLE was investigated in another cohort study 23 . Although the study indicated a significant increase in the mean QTc interval in the HCQ-treated group, performing a comprehensive multiple logistic regression analysis did not reveal significant disparities between the HCQ-treated and control groups.…”

“…A wide range of side effects such as cardiovascular, dermatological, digestive, hematological, metabolic, ophthalmologic, as well as other rare side effects were reported to be associated with HCQ use. 4 , 13 , 30 , 31 , 32 , 35 , 216 238 The main side effects of HCQ are summarized in Table 4 .…”
