QTL Mapping and Validation of Adult Plant Resistance to Stripe Rust in Chinese Wheat Landrace Humai 15

Abstract: Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst) is a devastating foliar disease that affects common wheat and barley throughout the world. The reasonable deployment of adult plant resistance (APR) wheat varieties is one of the best methods for controlling this disease. Wheat landraces are valuable resources for identifying the genes/QTLs responsible for disease resistance. Humai 15 is a Chinese spring wheat landrace and it has exhibited adequate levels of APR to the prevalent Pst races in field… Show more

“…In addition, structural changes in wheat chromosomes can be detected by FISH. For example, FISH was used to confirm a reciprocal translocation of the 5BL-7BS chromosome in a wheat variety Humai15 and a reciprocal translocation of the 4BL-6AL chromosome in a wheat variety Mingxian 169 [44]. In this study, however, FISH did not detect 5DL-5NsL.5NsS chromosome, therefore, 5NsL·5NsS might undergo another translocation within wheat chromosome, which is confirmed by FISH identification of two translocations, 3DS-5NsL·5NsS and 5DL-3DS·3DL in TR77.…”

“…IT numbers of 0, 1-3, 4-6, and 7-9 were considered immune, resistant, intermediate, and susceptible, respectively. DS was recorded as the percentage of the diseased leaf area [43,50]. For the F 5 plants, we selected resistant plants with full spikes, plant height less than 1 m, and a tiller number of 4 or greater.…”

“…Association analysis of stripe rust resistance using the general linear model + population structure (GLM + PC) SNP ID Chro. P-value Target allele (1) Allele effect (2) The left allele increased the resistance, (2) The effect of the left allele associated with increasing the resistance, (3) Phenotypic variation explained by marker. Number of markers signi cantly associated with the resistance to the Egyptian race of stripe rust at adult growth stage in 2019 and 2020 and the common markers between both years: a. signi cant SNP markers based on the GWAS using GLM+PC model, b. signi cant DArT markers based on single marker analysis (SMA).…”

“…Tritici), also known as yellow rust, is one of the most devastating wheat diseases in the world. It can cause losses in yield from 10 to 100% [1]. New stripe rust races appeared around the year 2000, spread rapidly, and caused signi cant yield losses [2,3].…”
